EPPF - Protein Production

Providing rapid and robust solutions for the production and purification of high quality protein reagents is critical for large number of projects across structural and translational biology. The protein production resources provided through EPPF are all located on the third floor of the Swann Building, with specialised areas including a purification suite with four ÄKTA Pure 25 systems and two ÄKTAXpress systems. ÄKTA Pure 25 is a new generation, flexible and intuitive chromatography systems, running UNICORN 7.1 control software, designed for fast purification of proteins, peptides, and nucleic acids from microgram to gram levels.




There is also a large core column library, a cell growth area and wet labs housing all the reagents and equipment equipment needed to produce high quality proteins.  The EPPF labs allow you to benefit not only from a centralised core laboratory, equipped with state-of-the-art equipment, but also from the dedicated team of  highly experienced technologists. They are responsible for the management of the laboratory, provision of technical assistance, training design of experiments and data interpretation.

Incubation/Cell Culture

  1. InFors Multitron - 6 cooling incubators for bacterial & yeast culture.


  1. InFors Multitron - 4 cooling incubator for insect cell culture.

Cell Lysis/Disruption

  1. Constant systems 1.1 kW TS cell disruptor - Automated high-pressure (40 kpsi) lysis system with temperature control. 2 ml – 20 litres extract handling capacity. Mammalian/insect/bacteria/yeast/plant origin samples.


  1. Soni-Prep 150 - Sonicator

Centrifugation

  1. Beckman Avanti 26i. Process – high-speed centrifugation (2 units). ~ 50 ml - 6 x 1 L.


  1. Beckman Allegra-X15 R - Small-scale ≤ 250 mls (12,000 xg) (2 units).

Purification

  1. ÄKTA Pure 25 systems - Automated scouting HPLC, for preparative/semi-analytical purification (4 units at 4˚C).


  1. ÄKTAXpress - Low pressure, automated, unattended parallel lab-scale production units (2 units at 4˚C).


  1. ÄKTAMicro - micro scale HPLC analytical chromatography/SEC-MALS.
