The EPPF has a large core column library, housed in a dedicated chiller cabinet in the purification suite. The library contains a wide variety of preparative columns and resins, as well as micro-preparative and high performance analytical columns.


There is redundancy for some of the most commonly used columns; see the on-line booking resource for details on the numbers of individual columns.


The EPPF also has a large selection of 1 and 5 ml prep-packed disposable columns (covering most preparative applications; e.g. IEX, HIC, AF, GF and HA). There is also a selection of loose resins and empty columns that the EPPF will pack for you. should you wish. Please speak to EPPF staff for more details on types and costs.


Please note that all columns should be re-equilibrated back into fresh 0.2 µm filtered ddH2O after use, unless otherwise instructed by EPPF staff.

Anion-exchange (AIEX):


EPPF-MiniQ PC 3.2/3 – 0.24 ml pre-packed     MiniQ PC 3.2/3 column for fast, sensitive and high- resolving cation exchange separations of proteins, peptides, oligonucleotides and other biomolecules in the micro-preparative scale. Max Back pressure 10 MPa. Rec flow rates 0.1 - 0.5 ml/min (max 1.0 ml/min). When running at a flow rate of 0.4 ml/min, the column generally operates at a back pressure of 4 MPa.). Mini Q beads are non-porous and have a particle size of 3 μm. The charged group on the gel is -CH2N+(CH3)3. Ionic capacity is 0.085 to 0.115 mmol/ml gel. Max loading capacity ~ 1 – 1.5 mg. Practical loading range 100 µg - 200μg.


EPPF-MonoQ 5/50 GL – 1 ml pre-packed glass columns for high performance anion ion exchange chromatography of proteins, peptides, polynucleotides and other biomolecules. Max Back pressure 4 MPa. Rec. flow rates 0.5  - 3 ml/min. Max loading capacity ~ 50 mg protein (sample dependent).


EPPF-Resource Q - 6 ml pre-packed strong anion ion exchange columns. Max Back pressure 0.6 MPa. Rec. flow rates 3 - 10 ml/min. Max loading capacity ~ 150 - 200 mg protein (sample dependent).


EPPF-Source 30Q 5/50 GL MAW – 1 ml glass column for intermediate stage anion ion exchange chromatography of proteins, peptides, polynucleotides and other biomolecules. (N/m = 8,000 m-1 / As = 1.2) Max Back pressure 1 MPa. Rec. flow rates 0.5 - 3 ml/min. Max loading capacity ~ 50 mg protein (sample dependent).


EPPF-HiPrep 16/10 Q FF - 20 ml pre-packed strong anion exchange column.  Max Back pressure 0.15 MPa. Rec. flow rates 2 - 10 ml/min. Max loading capacity ~ 100 mg protein/ml resin (sample dependent).


EPPF-HiPrep 16/10 DEAE FF – 20 ml pre-packed weak anion exchange column. Max Back pressure 0.15 MPa. Rec. flow rates 2 - 10 ml/min. Max loading capacity ~ 110 mg protein/ml resin (sample dependent).


Cation exchange (CIEX):

EPPF-HiPrep 16/10 SP FF - 20 ml pre-packed strong cation exchange column. Max Back pressure 0.15 MPa. Rec. flow rates 2 - 10 ml/min. Max loading capacity ~ 60 mg protein/ml resin (sample dependent). 


EPPF-HiPrep 16/10 CM FF – 20 ml pre-packed weak cation exchange column. Max Back pressure 0.15 MPa. Rec. flow rates 2 - 10 ml/min. Max loading capacity ~ 55 mg protein/ml resin (sample dependent).


Gel-filtration (GF):

EPPF XK16/40 Sephacryl S-400 HR – 69 ml (1.6 cm x 34.5 cm) in-house packed gel filtration columns designed for preparative purification of large protein complexes. (N/m ≥ 10,000 m-1, As = 0.87). Separation range (Mr) globular proteins 20 kDa. - 8 MDa. Max back pressure 0.2 MPa. Rec flow rate 0.2 - 1.2 ml/min (max = 2 ml.min-1). Max sample volume 1.4 ml. Recommended ≤ 1 ml for optimal resolution.


EPPF-HiPrep 26/10 Desalt - 50 ml pre-packed column for group separation/rapid desalt of high (Mr > 5,000 Da.) from low molecular weight substances (Mr < 1,000 Da.). Max Back Pressure 0.15 MPa. Rec flow rates 5 - 30 ml/min. Sample vol. 5 - 10 ml (15 ml max.)


EPPF-Superdex Peptide 10/300 GL – 24 ml pre-packed column for the high performance gel filtration of natural, recombinant or synthetic peptides and other small biomolecules. Separation range 100 - 7,000 Da. Max back pressure 1.8 MPa. Rec flow rates 0.2 - 1 ml/min. Max sample volume 500µl. Recommended ≥ 200 µl for optimal resolution.

Affinity (AF):


EPPF-HISPrep 16/10 FF - 20 ml prepacked, pre-charged Ni2+Sepharose™ 6 Fast Flow column for preparative purification of histidine-tagged recombinant proteins. Max Back pressure 0.15 MPa. Rec. flow rates 1 - 10 ml/min. Max loading capacity ~ 10 - 40 mg protein/ml resin (sample dependent).


EPPF-GSTPrep 16/10 FF - 20 ml prepacked Glutathione 4 Fast Flow column for preparative purification of GST-tagged recombinant proteins. Max Back pressure 0.15 MPa. Rec. flow rates 0.2 – 1.0 ml/min for loading, 1 - 10 ml/min for equilibration, wash and elution phases. Max loading capacity ~ 4 - 25 mg protein/ml resin (sample dependent).


Hydrophobic Interaction (HIC):


EPPF-HiPrep 16/10 Phenyl FF - 20 ml pre-packed column for hydrophobic interaction chromatography (HIC). Max Back pressure 0.15 MPa. Rec. flow rates 2 - 10 ml/min. Ligand density ~ 40 µmol/ml resin.


Chromatofocusing (CF):


EPPF-MonoP 5/50 GL - 1 ml pre-packed glass columns for high performance chromatofocusing of proteins, peptides, polynucleotides and other biomolecules with differences in pI down to 0.02 pH-units. Max Back pressure 4 MPa. Rec. flow rates 0.5 - 1.5 ml/min. Max loading capacity ~ 5 - 10 mg protein (sample dependent).


Reverse Phase (RPC):


NB! – these columns can only be used on the AKTA Ettan if fractionation is required.


SOURCE 15RPC ST 4.6/100 - 1.7 ml prepacked stainless steel column for fast, high-capacity preparative protein purification with reversed phase chromatography. Max Back pressure 4 MPa. Rec. flow rates 0.5 - 2.5 ml/min. Max loading capacity ~ 5 - 30 mg protein (sample dependent).


SOURCE 5RPC ST 4.6/150 – 2.5 ml prepacked, stainless steel columns for analytical and preparative reversed phase chromatography (RPC). Suitable for LC-MS applications. Max Back pressure 40 MPa. Rec. flow rates 0.1 - 1 ml/min. Max loading capacity ~ 5 - 30 mg protein (sample dependent).


µRPC C2/C18 ST 4.6/100 – 1.66 ml prepacked (3 µm porous silica beads with a mixture of C2 and C18 alkyl ligands) stainless steel column for fast, sensitive, and highest-resolution reversed phase chromatography at microscale. Suitable for LC-MS applications. Max Back pressure 35 MPa. Rec. flow rates 0.1 – 1.2 ml/min. Max loading capacity ~ 2 mg protein/peptide (sample dependent).
